Serien följer den excentriska läraren Ms. Frizzle, röstad av Lily Tomlin, som för sin klass på fantastiska utbildningsresor med hjälp av en magisk skolbuss. Eleverna i klassrummet är Carlos, Ralphie, Keesha och Wanda, röstade av Daniel DeSanto, Stuart Stone, Erica Luttrell respektive Lisa Yamanaka. Under äventyren reser gruppen till olika destinationer som yttre rymden, havets djup, myrstackar och även in i människokroppen.
Varje avsnitt kombinerar underhållning med faktabaserad undervisning om naturvetenskap, biologi och andra vetenskapliga ämnen. Serien presenterar komplexa vetenskapliga koncept på ett sätt som är tillgängligt för barn. Ms. Frizzles motto "Seat belts, everyone!" blev ikonisk för serien och markerade början på varje nytt äventyr.
