I varje avsnitt besöker familjen nya platser och möter olika situationer som lär dem nya koncept och idéer. Serien kombinerar lärande med underhållning genom att låta karaktärerna upptäcka världen omkring sig under sina resor. På båten finns även en femte karaktär, Peekaboo, som är mer tillbakadragen och följer med i äventyrena utan att de andra Twirlywoos alltid märker det.
Serien presenterar också återkommande gäster som besöker båten, inklusive Stop Go Car och The Very Important Lady. Dessa möten skapar nya situationer och lärandemöjligheter för huvudkaraktärerna. Twirlywoos är utformad för att engagera små barn genom färgglada animationer, enkla narrativ och fokus på utforskning och upptäckter.
